Abstract
Considerable evidence suggests that during the progression of complex diseases, the deteriorations are not necessarily smooth but are abrupt, and may cause a critical transition from one state to another at a tipping point. Here, we develop a model-free method to detect early-warning signals of such critical transitions (or un-occurred diseases), even with only a small number of samples. Specifically, we theoretically derive an index based on a dynamical network biomarker (DNB) that serves as a general early-warning signal indicating an imminent sudden deterioration before the critical transition occurs. Based on theoretical analyses, we show that predicting a sudden transition from small samples is achievable provided that there are a large number of measurements for each sample, e.g., high-throughput data. We employ gene expression data of three diseases to demonstrate the effectiveness of our method. The relevance of DNBs with the diseases was also validated by related experimental data (e.g., liver cancer, lung injury, influenza, type-2 diabetes) and functional analysis. DNB can also be used for the analysis of nonlinear biological processes, e.g., cell differentiation process.
个人简介:陈洛南,1984年获华中科技大学电气工程学士学位;1988年获日本东北大学系统科学硕士学位;1991年获日本东北大学系统科学博士学位。1997年起任日本大阪产业大学副教授;2000年起任美国加州大学洛杉矶分校(UCLA)访问教授;2002年起任日本大阪产业大学教授;2009年4月起任日本东京大学教授(兼);2009年10月至今任中科院系统生物学重点实验室执行主任,研究员。现任中国运筹学会《计算系统生物学分会》理事长,IEEE-SMC《系统生物学委员会》主席,中日韩国际系统生物学会组织(Trisys)的轮值主席,中国细胞生物学会《功能基因组学与系统生物学分会》副会长,中国药理学会《网络药理学专业委员会》副主任委员,上海市临床生物信息学研究所副所长,国家基金委重大研究计划“高性能科学计算的基础算法与可计算建模”专家组专家,国家重点研发计划首席科学家。主要从事计算系统生物学和大数据分析的研究工作。近年来,陈洛南在系统生物学等研究领域发表了300篇以上SCI期刊论文及10部以上编著书籍。

sexually transmitted diseases on the coupled network

Abstract
Sexually transmitted infections (STIs) have caused significant public health burden globally. In most models of STIs spreading in networks, it is assumed that the transmission and recovery processes are Markovian processes; that is, the transmission and recovery times per event are exponentially distributed. However, both transmission and recovery times per event for many real STIs do not completely follow exponential distribution, which leads to disagreements, both qualitatively and quantitatively, with models under the exponential distribution assumption. In this talk, allowing for general transmission and recovery times distributions, we propose a general Edge-Based Age-structured-like Compartmental Model for STIs (EBACMS) in a coupled configuration network. We consider sexual transmissions between men with also heterosexual contacts. The obtained system is a coupled system of ordinary differential equations and partial differential equations. We study the existence and non-negativity of solutions, the epidemic threshold R0, the global stability of disease-free equilibrium with R01. Sensitivity analysis on R0 and F in terms of the parameters illustrates that male-to-male routes have a greater impact on R0 and F than the heterosexual transmission routes when the transmission process is Markovian and the recovery process is arbitrary.
